“台农甜蜜”桃胚愈伤组织的诱导与继代保持 被引量:3

摘要 以5年生"台农甜蜜"桃接近成熟的胚为试验材料,研究接种方式、光照条件和不同培养基对其愈伤组织诱导影响。结果表明,黑暗条件下,将去除种皮的"台农甜蜜"桃子叶胚接种到附加2.0mg/L的MS培养基上培养,可以诱导出松散的、生长状态良好的桃愈伤组织。在"台农甜蜜"桃愈伤组织继代保持过程中,比较了光照条件、愈伤组织类型、不同培养基和继代方式对其生长的影响。结果表明,采用附加1.0mg/L2,4-D的MS培养基与附加1.0mg/L2,4-D、0.5mg/LKT、100mg肌醇的MS培养基交替继代,并在光照下培养,可以实现"台农甜蜜"桃TN2和TN3类型愈伤组织的长期继代保持,并使其保持旺盛的生长能力。 The mature embryos of 5 old years' cultivars of 'Premier' honey peach were used as the materials in this paper. The effects of the inoculation method, light treatment and different mediums on the callus induction were studied. The results showed that the friable and well callus were obtained by the embryos without spermoderm on the MS with 2.0mg/L 2,4-D under the dark. The comparisons of the effects of the light treatment, callus type, different mediums and subcuhuring method on the growth of the honey peach callus were investigated in the process of their subculture and maintenance. The conclusions were summarized as follows, the TN2 and TN3 of the honey peach callus could be maintained for a long time and kept exuberant growth when they were alternately cultured on the MS medium containing with 1.0 mg/L 2,4-D and the other MS medium supplemented with 1.0 mg/L 2,4-D, 0.5 mg/L KT and 100 mg/L inositol.
