摘要
为有效利用烟草资源,建立一种快速提取烟碱的方法,以烟草为原料,采用超声波法提取烟碱,气相色谱法测定,通过单因素试验分别研究不同甲醇浓度、超声时间、料液比对烟草烟碱提取量的影响,试验结果表明,超声波法提取烟草烟碱具有操作简单、快速的优点,单因素试验结果:当甲醇浓度为40%时,烟碱的提取量可达(41.52±0.28)μg/g;超声时间为45min,烟碱提取量可达(39.92±0.25)μg/g;料液比为1:20,烟碱提取量可达(42.21±0.16)μg/g。并通过三元一次回归正交设计方法,研究了甲醇浓度(x1)、超声时间(x2)、料液比(x3)与烟碱提取量(y)之间的关系,得到了烟碱提取量(y)和提取条件的回归方程:y=35.930-2.788x1+0.842x2-0.985x3-1.278x1x3(10%≤x1≤70%,15min≤x2≤45min,1:10≤x3≤1:30);R2=0.968,为进一步优化提取条件提供了一定的理论依据。
In order to use resources on tobacco effectively, and establish a rapid extraction method of nicotine. This test used tobacco as raw materials, methanol as impregnate, extracted nicotine in tobacco by ultrasonic wave, and then determined nicotine by gas chromatography. Studied effect of different concentrations of methanol, ultrasonic time and the ratios of material to liquor on quantity of nicotine in tobacco. The results show that extract nicotine in tobacco by Ultrasonic, this method is very easy and quick .When concentration of methanol was 40%, the quantity of nicotine was (41.52±0.28) μg/g; when ultrasonic time was 45 min, the quantity of nicotine was (39.92±0.25) μg/g; when the ratio of material to liquor was 1:20, the quantity of nicotine was (42.21±0.16)μg/g .And the also studied relationship of different concentrations of methanol (x1), ultrasonic time (x2), the ratios of material to liquor (x3)and the quantity of nicotine (y)by using method of multiple regression orthogonal design. Got the regression equation of extraction quantity of nicotine and different conditions: y=35.930-2.788x1+0.842x2-0.985x3-1.278x1x3 (10%≤x≤70%, 15min≤x2≤45min, 1: 10≤x3≤1:30) R^2=0.968.
