摘要
目的观察经冠状静脉窦逆行灌注血管内皮生长因子基因(pAdtrackCMV-VEGF165)对其表达分布的影响。方法20只新西兰大白兔随机分为实验组(VEGF基因治疗组)和对照组,2组动物常规全麻,气管插管,胸骨正中切开,暴露心脏,结扎左室支建立心肌梗死模型,冠状静脉窦插管,实验组逆行灌注pAdtrackCMV-VEGF165,对照组以生理盐水对照。全程记录心电,1周后取梗死区(左室心尖部)及非梗死区(高侧壁)心肌,采用逆转录聚合酶链反应(RT-PCR)检测基因表达情况。结果对照组1只动物死亡,实验组所有动物全部存活;结扎左室支即刻所有动物出现ST段抬高并与T波融合;RT-PCR显示实验组梗死区检测到转染基因表达,而非梗死区及对照组则未检测到转染基因表达。结论冠状静脉窦逆行灌注技术能很好地将VEGF基因靶向转移至梗死区。
Objective To observe the affection of distribution and expression of vascular endothelial growth factor gene (pAdtrackCMV-VEGF165) by coronary sinus retrograde perfusion in rabbit myocardial infarction model Methods 20 New Zealand rabbits were divided into 2 groups which were experimental group and control group. 2 groups animals were general anesthesia and open thorax, elevating the apex of the heart, clamping the main stem of left ventricular coronary , pAdtrackCMVVEGF165 were retrograde perfused trough coronary sinus in experiment group. The physiological sa- line was used in control group. The distribution and expression of pAdtrack CMV-VEGF165 were detected 1 week later at different point of left vetrencle. Results 1 animal died in control group and all an- imal survived in experimental group. The ST segment of animals elevated immediately and fused with T wave after the ligation of the left main ventricular branch. In experiment group, the expression of transferred gene can be detected in infarction area by using RT-PCR technique. However, the expression of transferred gene was not detected in non-infarction area as well as in control group. Conclusion Retrograde coronary sinus perfusion technique could perfectly distribute VEGF gene to the target ischemia area in rabbit myocardial infarction model.
