摘要
目的建立液质联用(HPLC—MS/MS)法同时测定人全血中环孢素A、他充莫司、西罗莫司的方法。方法采用Symmetry C18色谱柱(2.1mm×50mm,5μm),流动相:10mmol·l^-1醋酸铵-0.1%甲酸溶液和0.1%甲酸甲醇溶液为90:10,流速0.25mL·min^-1,采用多反应监测进行定量.结果环孢素A在人全血中50~1500μg·L^-1。内线性良好,他克莫司、西罗莫司在人全血中1~30μg·L^-1内线性良好,相对回收率在98.67%~103.56%之间,日内及日间精密度(PSD%)均小于10%。结论本方法能满足同时测定人全血中环孢素A、他克莫司、西罗莫司的要求,用于临床血药浓度的监测。
OBJECTIVE To develop a HPLC/MS/MS method for simuhanenus determination of cyelosporin A, tacrolimus and sirolimus in human blood. METHODS The separation was performed on a Symmetry C18 (2. 1 mm × 150 mm,5 μm) column with a mobile phase of water (containing 10 mmol· L^-1 ammonium acetate and 0. 1% formic acid) and methanol (containing 0. 1% formic acid). Multi-reaction monitoring mode was employed for the quantitative determination. RESULTS The standard curve of cyclosporine was linear over the range of 50 - 1 500 μg · L^-1. The standard curves of tacrolimus and sirolimus were linear over the range of 1 - 30 μg · L^-1 , the relative recovel7 was among the range of 98.67% - 103.56% , RSDs of intraand inter-day validation were less than 10%. CONCLUSION HPLC-MS/MS can be nsed for the simultaneous determination of cyclosporin, tacrolimus and sirulimus in human blood.
