一株降解DON毒素真菌的鉴定及其生理特性研究 被引量:1

摘要 为了对实验室已分离的1株高效降解脱氧雪腐镰刀菌烯醇毒素(DON)的真菌(NJA-1)鉴定,对该菌进行了形态学观察以及rDNA ITS区基因序列的扩增、测定。形态学观察发现该菌属于曲霉属黑色组曲霉黑曲霉集合体。进一步分别用引物对BMB-CR和ITS2及ITS1和ITS4扩增rDNA的ITSⅠ-5.8S rDNA-ITSⅡ,得到总长为738 bp的基因序列,与GENBank中已有的基因序列比对和亲缘关系比较分析发现该菌rDNA ITS区基因序列与塔宾曲霉的同源性为100%,最终确定NJA-1为1株塔宾曲霉。菌株NJA-1 rDNA ITS区基因序列收录入GENBank,登陆号为EF178271。还对NJA-1的生长特性、炭源和氮源的利用、适宜生长的培养基pH等生理特性进行了测定,为该菌的大量培养提供数据基础。 Morphological observation and rDNA ITS domain sequence amplification and measurement were carried out in order to identify a DON (deoxynivalenol) degradable fungal strain (NJA-1). It was found morphologically that the fungus belongs to genus of AspergiUus niger aggregate. Later a total length of 738 bp gene sequence was obtained respectively using primer to amplify ITSⅠ-5.8S rDNA-ITS Ⅱ of rDNA for BMB-CR, ITS2 and ITS4. And it was compared with existing sequence in GENBank and relationship comparison analysis found that rDNA ITS of the strain had 100% of homology with A. tubingensis. Finally NJA-1 was confirmed as A. tubingensis. And rDNA ITS domain gene sequence was collected by GENBank (No. EF178271 ). Its growth characteristics, utilization of carbon and nitrogen sources, suitable culture medium, pH, and physiological features were also studied and measured thus laid data foundation for culture the strain in large scale.
