Determination of Sperm DNA Fragmentation in Infertile Males by Sperm Chromatin Dispersion Test

摘要 目的:通过精子染色质扩散(SCD)实验检测精子DNA碎片,探讨男性不育患者精子DNA完整性。方法:改进的SCD实验分析精子DNA碎片,DNA完整的精子产生扩散的大光晕或中光晕,而DNA损伤的精子不产生或产生很小的光晕。少弱精子症患者56例,畸形精子症不育患者31例,对照组32例。结果:对照组、少弱精症组和畸形精子症组大、中光晕精子比率分别平均为(88.0±5.9)%、(73.4±9.6)%和(58.5±11.2)%,精子DNA碎片比率分别平均为(12.0±5.2)%、(26.6±9.7)%和(41.5±12.3)%,少弱精症组和畸形精子症组与对照组比较有统计学意义(DNA完整的精子比率显著低于对照组,P<0.001,精子DNA碎片比率显著高于对照组,P<0.001)。结论:SCD实验是检测精子DNA完整性有效的方法。少弱畸精子症不育患者精子DNA碎片比率增高。 Objective: To determine the sperm DNA fragmentation by sperm chromatin dispersion (SCD) test, and explore the sperm DNA integrity of infertile males. Methods: The sperm DNA fragmentation was determined by improved SCD test in infertile males with oligoasthenozoospermia (n=56) and teratozoospermia (n=31), and 32 adult healthy fertile males were served as the control group. Sperm nuclei with integrity DNA produced large or medium-sized dispersion halos, while nuclei with DNA fragmentation produced very small or no halos. Results: In the control group, the oligoasthenozoospermia group and the teratozoospermia group, the mean percentages of sperm nuclei with large and medium-sized halos were respective (88.0±5.9)%, (73.4±9.6)% and (58.5±11.2)%, and with small or no halos were (12.0±5.2)%, (26.6±9.7)% and (41.5 ±12.3)%, respectively. There were statistical differences between the infertile groups and the control group: the percentages of sperm with integrity DNA of the infertile groups were significantly lower than the control group (P〈0.001), and the percentages of DNA fragmentation were significantly higher than the control group (P〈0.001). Conclusions: The SCD test is an effective method for determining sperm DNA integrity. The ratio of sperm DNA fragmentation increases in infertile males with otigoastheno-and teratozoospermia.
