摘要
目的:由于转移因子口服冻干粉制剂中的辅料甘露醇会对福林酚法的测定产生干扰,因此对甘露醇对测定方法的干扰进行研究,以期在实际测定中可予以预测并排除;方法:以牛血清白蛋白为对照,制备标准品并得出标准曲线,对比不含甘露醇和含0.27%甘露醇转移因子溶液的吸光度;结果:0.27%甘露醇福林酚溶液在650nm处有吸收峰,因此可能影响多肽的测定结果,不同批号的甘露醇与福林酚试剂的呈色反应不一,吸光度值最多可差4倍以上;结论:测定管减去空白管(福林酚0.27%甘露醇)的吸光度,再计算,可得到准确的实验结果。在检测以甘露醇为辅料的转移因子冻干制剂时,空白管必须要采用同批的甘露醇。
Objective :To study and exclude the interruption of mannitol on the detection of peptide in transfer factor by Folin-reagent method. Methods:Use bovine serum albumin as standard control to get standard curve. Compare the OD650nm of transfer factor solution with mannitol to that of without mannitol. Results:There is an absorptive peak at 650nm of 0.27% mannitol-Folin solution and it may interrupt the result of peptide detection. The reaction and OD650nm differs a lot. Conclusion:It is necessary to subtract the OD650nm of 0.27% mannitol-Folin solution and use mannitol with the same Batch number.
