摘要 目的观察熊果酸(ursolic acid,UA)对脑胶质瘤U251细胞株增殖及细胞外信号调节激酶(extracellularsignal-regulated kinase,ERK)、细胞周期蛋白(cyclin)D1/p21waf/cip1信号通路的影响。方法培养脑胶质瘤细胞株(U251),四甲基偶氮唑蓝(MTT)比色法、克隆形成实验及流式细胞术测定不同浓度UA对细胞增殖的影响;Western-blot测定p-ERK1/2及下游的cyclin D1、p21waf/cip1表达。结果MTT、克隆形成实验及流式细胞术均显示UA可明显抑制U251细胞增殖,免疫印记法显示UA可明显抑制p-ERK1/2、cyclin D1表达及增强p21waf/cip1表达。结论UA可抑制脑胶质瘤U251细胞株增殖,其途径与下调p-ERK1/2表达、增强p21waf/cip1表达进而抑制cyclin D1表达有关。 Objective To investigate the effects of ursolic acid ( UA ) on the proliferation of glioma cell U251 and signal pathway as ERK and cyclin D1/p21^waf/cip1 Methods The effect of UA on proliferation of cell line U251 were observed by MTTssay, clone formation experiment and/low cytometry; p-ERK1/2,cyclin Dl and p21 ^waf/cip1 expressions were assessed by Western blot. Results The proliferation of cell line U251 was significantly inhibited, intracellular p-ERK1/2 and cyclin DI expression were suppressed, while p21 ^waf/cip1 expression was enhanced by different concentrations of UA. Conclusion UA decreases the proliferation of cell line U251 in the dose dependent manner, which may be due to down-regnlation of ERK, upregulation of cyclin Dl/p21^waf/cip1 and followed cyclin D1suppression.
