摘要
目的探讨"派莎"防晒乳膏对中波紫外线(UVB)损伤人角质形成细胞(HKC)的保护作用及作用机制。方法用"派莎"防晒乳膏对HKC进行预处理24 h,采用不同强度的UVB照射体外培养的HKC,24 h后以噻唑蓝(MTT)法检测细胞生存率,用酶联免疫吸附(ELISA)法检测上清液中白细胞介素(IL-8)浓度,以酶联免疫吸附实验检测细胞上清液中肿瘤坏死因子(TNF)-α的分泌量。结果经UVB照射后,细胞损伤程度与UVB照射剂量有关,"派莎"防晒乳膏可提高UVB照射后HKC的生存率,减少细胞因子TNF-α的分泌。UVB照射可使HKC分泌IL-8增加,"派莎"防晒乳膏组IL-8浓度与对照组比较差异有显著性(P<0.05)。结论UVB对HKC有损伤作用,且与照射强度相关,"派莎"防晒乳膏对HKC具有光保护作用。抑制TNF-α和IL-8的释放,提高UVB照射后HKC的生存率,可能是其保护作用机制之一。
Objective To investigate the photo-protective effect of paisha sunscreen cream on human keratinocytes exposed in UVB radiation. Methods After pretreated with paisha sunscreen cream for 24 hours, HKC cells were irradiated with different dosages of UVB. The cell viability was detected by MTT method, The snpernatant was collected at 24 hours post irradiation to measure the secretion of cytokine TNF-α and IL-8 by ELISA. Results The cell viability was inverse correlated with the irradiated dosage. Paisha sunscreen cream could boost the cell viability, reduce the secretion of TNF-α. The secretion of IL-8 was increased after irradiation by UVB. There was a significant difference of IL-8 level between paisha sunscreen cream groups and the control group (P 〈 0. 05). Conclusion The damage of UVB irradiation on human keratinocyte was dose-dependent. Paisha sunscreen cream played a role of photo-protection through inhibition of TNF-α and IL -8 secretion and improvement of cell viability.
