摘要
研究近红外漫反射光谱(NIRDRS)同偏最小二乘法(PLS)相结合对四种碱基(鸟嘌呤G、腺嘌呤A、胞嘧啶C、胸腺嘧啶T)混合物进行定量分析。实验结果表明:当最佳主成分数为4时,碱基含量的预测值最好,其预测集的相关系数R分别为A=0.9847,T=0.9851,G=0.9923,C=0.9933;相对标准误差RSD分别为A=1.3927%,T=1.4952%,G=1.6353%,C=1.7092%。
The quantitative analysis of DNA base mixture (A, G, C, T) was made by Near-infrared Diffuse Reflectance Spectrometry (NIRDRS) and Partial Least Squares (PLS). The results showed that the best predictions of base contents were occurred when optimum main ingredient equaled 4. The correlative coefficient of prediction set were 0.9847 (A), 0.9851 (T), 0.9923(G), 0.9933(C) respectively. Relative standard deviations of four bases were 1.3927 % (A), 1.4952% (T), 1.6353% (G) and 1.7092%(C) respectively.
