摘要
目的建立毛细管电泳方法用于测定麻黄汤中的麻黄碱、伪麻黄碱和苦杏仁苷的含量。方法:采用熔融石英毛细管柱(id:75cm×75μm,有效长度66.5cm),缓冲体系:25mmol.L-1硼砂缓冲液(pH=9.3),工作电压为20kV,柱温28℃,波长检测194nm,对麻黄汤中的麻黄碱、伪麻黄碱和苦杏仁苷3种有效成分进行定量分析。结果麻黄碱、伪麻黄碱和苦杏仁苷为基线分离;平均回收率分别为98.82%、105.16%、96.31%;RSD分别为1.94%、2.03%、1.78%。回归方程和相关系数分别为:y=98.17x+9.95,r=0.9994;y=232.17x+113.41,r=0.9966;y=745.19x+182.65,r=0.9990。结论此法简捷、快速、灵敏,可用于麻黄碱、伪麻黄碱和苦杏仁苷的定量分析。
AIM To set up a simple,fast method of capillary electrophoresis to analyze the content of ephedrine,pseudoephedrine and amygdalin of Ephedra Decoction.METHODS The ephedrine,pseudoephedrine and amygdalin were detected at 194 nm by high performance capillary electrophoresis(HPCE).The optimized condition was 25 mmol·L-1 boronic acid buffer with pH=9.3.The working voltage was 20 kV,the temperature was 28℃,and the capillary was 75 cm×75 μm(effective length 66.5 cm).RESULTS The three effective ingredients were separated baselinely.The average recoveries for ephedrine,pseudoephedrine and amygdalin were 98.82%,105.16%,96.31% respectively,with the RSD 1.94%,2.03%,1.78%.Their regression equations were y=98.17x+9.95,y=232.17x+113.41,y=745.19x+182.65 respectively.CONCLUSION This method is simple,quick and sensitive.It can be used to control the quality of Ephedra Decoction
