腺病毒受体在不同白血病细胞中的表达及意义

摘要 高效转移目的基因进入细胞,尤其是处于非分裂静止期的细胞,是基因治疗至关重要的步骤。由于腺病毒载体(AdVec)具有制备简便、宿主范围广、感染性强、包装容量大、病毒繁殖滴度高和安全性好的特点,因此,AdVec已成为继逆转录病毒载体后被广泛应用的载体系统。然而,我们以往的研究发现,传统的以5型腺病毒为基础构建的AdVec即Ad5Vec很难高效感染多种不同的白血病细胞如K562细胞,但是,它们均能被新型嵌合的AdVec即Ad5F35AdVec有效感染。为进一步探讨其机制,我们分析了腺病毒受体(CAR)在不同白血病细胞中的表达。
