刺孢小克银汉霉产油脂的提取方法研究

摘要 以刺孢小克银汉霉为载体,在分离有机溶剂的方法、不同溶媒、溶媒配比、酸热时间和次数方面对提取油脂的传统酸热法进行了改进研究,获得了新的酸热法,并将其与索氏提取法进行了含油率和气相色谱后成分的比较分析.新的酸热法,采用抽滤分液和正己烷、乙醇按2∶1混合萃取,两次热冷破壁.与传统的酸热法相比较,更为安全,简便,省去了离心的过程,油脂得率提高了20.71%;与索氏提取法进行比较:新酸热法单位时间处理能力是索氏提取法的96倍,设备要求低.它的油脂得率能达到索氏提取法的95%以上,GLA的含量较索氏提取法高出了17.45%,是一种真正高效、快速的能运用于工业化的提取方法. Using Cunninghamella echinulata as a carrier, a new acid heating extraction is acquired from improving traditional acid heating extraction in separating organic solvent, different solvent, the ratio of solvent, times of acid heating and degrees. A comparison is made between this new method and Soxhlet extraction in oil containing ratio and the components are analyzed after gas chromatography. By adopting extraction and filtration for dividing liquid and the mix ratio of n hexane and ethanol was 2.1 to extract as well as added two times of heating and cooling to damage cell wall, this new approach, leaving out centrifugal process and with the percent of acquired lipid increased by 20.71%, was safer, simpler and more convenient compared with the conventional way. Its dealing capability per hour was 96 times as much as that of Soxhlet extraction and the requirement of equipments was also low correspondingly when compared with the latter. Its attainment of lipid achieved 95% of that in Soxhlet extraction and the containment of GLA increased by 17.45% so that it is a new method that can be applied to industrialization because of its high effectiveness and rapid reaction.
