舒林酸抑制胶质瘤细胞株C6增殖的实验研究

摘要 目的观察非甾体类抗炎药舒林酸对胶质瘤C6细胞增殖的影响,并探讨其作用机制。方法采用四氮唑蓝(MTT)比色法观察舒林酸对C6细胞增殖的影响,采用流式细胞仪检测舒林酸对C6细胞凋亡和细胞周期分布的影响。结果舒林酸可抑制C6细胞增殖,可诱导细胞凋亡和G0/G1期阻滞,且呈现明显的时间和剂量依赖性(P<0.05)。结论舒林酸可抑制C6细胞增殖,其机制涉及影响细胞周期和诱导细胞凋亡方面。 Objective To explore the effects of Sulindac on the proliferation of human glioma C6 cells and its mechanisms. Methods The inhibitory effect of sulindae on proliferation of glioma C6 cells in vitro was determined by methyl thiazolyl tetrazoliun (MTT) method. The effects of sulindac on cell apoptosis and cell cycles phase distribution of human glioma C6 cells were measured by flow cytometry assays. Results The proliferation of human glioma C6 cells was significant inhibited by sulindac in vitro. Glioma C6 cells apoptosis and the increase in percentage of the cells during the G0/G1 phase were induced by sulindac. The above-mentioned effects were significantly time- and dose-dependent (P〈0.05). Conclusion Glioma C6 cell line can be inhibited by sulindae in vitro and its mechanism may be related to the change in cell cycle phase distribution and increase in cell apoptosis.
