摘要
目的:观察"中风1号"喷雾剂鼻腔给药对脑梗死大鼠急性期的治疗作用,以及其量效关系。方法:采用Longa线栓法制备大脑中动脉阻塞(MCAO)脑缺血模型大鼠,设模型对照组(A组)、"中风1号"剂量递增组(B组)(剂量从1∶100至1∶1)、中风1号1∶1剂量组(C组)。用药24h后观察大鼠一般状况和神经行为学、脑缺血梗死体积及局部脑血流量、运动血细胞数量以及局部血流速度的变化。结果:中风1号组对脑缺血模型大鼠24h内死亡率低于模型对照组;对脑缺血大鼠神经缺损功能的影响优于模型对照组;对脑缺血模型脑梗死体积的影响小于模型对照组。对局部脑血流的影响:B组用1∶100剂量的中风1号鼻腔给药,对缺血局部的脑血流量的变化无明显影响;用1∶20剂量有明显的升高局部脑血流量的作用,主要是增加运动血细胞数量;增加浓度至1∶2,升高局部脑血流量的作用达到最佳,主要是增加局部运动血细胞数量的作用依剂量递增。C组在局部血流速度有进一步增加的同时,运动血细胞数量却有下降,局部血流量不能进一步升高,反而较1∶2剂量有下降。结论:中风1号方能降低脑缺血模型大鼠24h内死亡率,减少脑缺血模型脑梗死体积,改善脑缺血大鼠神经缺损功能;中风1号在浓度为1∶2时,升高局部脑血流量的作用达到最佳。
Objective: To investigate the therapeutic effects of "Zhongfeng No. 1" Spray administered through nasal cavity on acute cerebral ischemia in rats and the dose-effect relationship of the drug. Methods: The cerebral ischemia model of middle cerebral artery occlusion (MCAO) was established with Longa suture method in rats. Three groups were set up in the study, they were modern control group(A group), "Zhongfeng No. l"dose-inerease group (B group) (doses from 1 : 100-1 : 1), and "Zhongfeng No.1" of concentration 1:1 group (C group). After 24 hours of administration, the general condition, neural behavior, Ischemic cerebral infarction volume, regional eerebral blood flow, number of moving blood cells and the changes of local blood flow velocity were detected respectively. Results: The mortality rates within 24 hours of B and C groups were lower than that of the control group; the degree of neural defect in B and C groups was less serious than that of the control group; the volume of cerebral infarction in B and C groups was smaller than that of control group. In the observation of regional cerebral blood flow, "ZHongfeng No.l" spray with concentration of 1:100 had no obvious affect on cerebral blood flow of the regional ischemia section. Increasing concentration of "Zhongfeng No.l" spray to 1:20 could significantly elevate, mainly increase the number of moving blood cells. When the concentration was increased to 1:2, the effect of "Zhongfeng No.l" spray on elevating the regional cerebral blood flow was the best of all. From 1:100 to 1:2, the effect of increasing the quantity of moving blood cells was enhanced with the increase of concentration. However, when the concentration was increased to 1:1, although the regional blood flow velocity increased, the number of moving blood cells decreased, leading to the the re- gional cerebral blood flow volume falling down, even lower than that of 1:2. Conclusion: "Zhongfeng No.l" spray can reduce the mortality within 24 hours of cerebral ischemia, diminish the volume of ischemic cerebral infarction, and alleviate the neural defect caused by the cerebral blood flow decrease. When the the concentration reaches 1:2, "Zhongfeng No.l" spray begins to show obvious effect of lifting the regional cerebral blood flow volume, and the best effect can be acquired at the concentration of 1:2.
