紫杉醇是治疗乳腺癌的抗微管解聚药。本文探讨紫杉醇对巨噬细胞分泌IL-6和TNF-α的影响,作为紫杉醇抗癌效应的另一机制。方法:正常鼠和乳腺癌瘤鼠腹腔巨噬细胞(PEM)与不同浓度的紫杉醇和大肠杆菌脂多糖(LPS)分别或混合培养。在24h和48h用ELISA及Western blot分析PEM分泌IL-6和TNF-α的变化。结果:用紫杉醇处理瘤鼠PEM24h,IL-6水平明显降低,与正常鼠PEM比较有非常显著性差异(P<0.01)。紫杉醇对PEM分泌IL-6的抑制作用可以被LPS拮抗。紫杉醇和LPS均能增强正常和瘤鼠PEM分泌TNF-α,两组间TNF-α水平有非常显著性差异(P<0.001)。Western blot表明紫杉醇能促进瘤鼠PEM的TNF-α合成。结论:紫杉醇促进瘤鼠PEM的TNF-α分泌和抑制IL-6的产生,佐以提高紫杉醇的杀瘤效应。
Aims:To clarify the characterization of the ability of Taxol to initiate macrophage tumor cell and to regulate IL-6 and TNF-** secretion. Methods: Normal and D-DMBA-3 murine with peritoneal exudate macrophages (PEM) were cultured with Taxol or LPS alone, or with combinations of Taxol and LPS. IL-6 and TNF-** levels were determined by the means of ELISA and Western blot at 24h and 48h, respectively. Results: Excretion of IL-6 was decreased significantly by treatment with Taxol at 24h. However, Taxol which inhibited the excretion of IL-6 by PEM can be blocked by LPS. Taxol or LPS weakly induced normal PEM to release TNF-**, and TNF-** levels of tumoral PEM were significantly higher than those of normal PEM (P<0.001). Taxol has been shown to activate tumoral PEM to express TNF-** by Western blot. Conclusions: This study demonstrates that Taxol clearly exhibits a strong influence on macrophage and may identify other way of Taxol antitumor effects.
