摘要
目的:检测表皮生长因子(EGF)对A549肺腺癌细胞黏附和侵袭能力的影响。方法:A549肺腺癌细胞分为对照组、1ng/mL EGF组和10ng/mL EGF组,常规培养48h,采用Fn包被法和Transwell小室法分别对3组细胞黏附和侵袭能力进行检测,分别以细胞黏附率和穿膜细胞数来表示细胞黏附和侵袭能力的大小。结果:EGF作用48h后,1ng/mL EGF组和10ng/mL EGF组A549肺腺癌细胞的黏附率和穿膜细胞数均显著高于对照组(P<0.01),但1ng/mL EGF组和10ng/mL EGF组A549肺腺癌细胞之间相比,细胞黏附率和穿膜细胞数无显著性差异(P>0.05)。结论:EGF可增加A549肺腺癌细胞的黏附和侵袭能力,但其黏附和侵袭能力并不随EGF浓度的增加而继续增加,提示EGF在正常浓度范围时即可显著促进A549肺腺癌细胞的黏附和侵袭。
Objective:To determine the influence of epidermal growth factor(EGF) on the adhensive and invasive ability of A549 lung adenoearcinoma cells. Methods: A549 lung adenocareinoma cells were divided into 3 groups ,including control group, 1ng/mL EGF group and 10ng/mL EGF group, then were routinely incubated for 48 hours. Fibronectin coating method and Transwell chamber method were employed to determine the adhensive and invasive ability of ceils in 3 groups respectively. The cell adhesive rates and cell counts passed through membrane were used to demonstrate the adhesive ability and invasive ability of ceils. Results:After incubation for 48 hours with EGF, the adhesive rates and cell counts passed through membrane in 1ng/mL EGF group and 10ng/mL EGF group were both significantly higher than that of control group ( P 〈 0. 01 ), but the adhesive rates and cell counts passed through membrane were not significantly different between 1ng/mL EGF group and 10ng/mL EGF group(P 〉0. 05). Conclusions:EGF can increase the adhesive ability and invasive ability of A549 lung adenocarcinoma cells ,but they are not continuously increased with the increasing concentration of EGF, suggesting that EGF at normal level can significantly improve the adhesion and invasion of A549 lung adenocarcinoma cells.
