摘要
[目的]建立简便、准确、灵敏的测定大蒜中果糖、葡萄糖及蔗糖含量的方法。[方法]样品经热水提取后,使用Kromasil NH2(250mm×4.6mm,5μm)柱分离,以乙腈-水(80:20,V/V)为流动相,使用蒸发光散射检测器检测。[结果]果糖、葡萄糖和蔗糖的线性范围分别为0.05~3、0.075-5、0.05~5g/L;检出限分别为1.9、2.4、1.8μg/L;样品的平均加标回收率分别为99.2%、97.6%、98.0%;相对标准偏差分别为1.02%、1.41%、1.27%。[结论]该方法操作简便、快速、准确,用于实际样品的测定,结果令人满意。
[ Objective ] The aim was to develop a simple, accurate and sensitive method for determination of carbohydrates in Allium sativum L. [ Method] The carbohydrates in Allium sativum L. were extracted with hot water and separated on Kromasil NH2 column(250 mm× 4.6 mm, 5μm) using acetonitrile and water as the mobile phase with volume ratio of 80:20. The elute from the column was examined by an evaporative light scattering detector. [ Result ] The linear ranges of fructose, glucose and sucrose were 0.05 - 3, 0.075 - 5 and 0.05 - 5 g/L. The detection limits were 1.9, 2.4, and 1.8 μg/L. The average recovery were 99.2%, 97.6% and 98.0%, and the relative standard deviation were 1.02%, 1.41% and 1.27%, respectively. [ Conclusion] The method was applied to the determination of carbohydrates in Allium sativum L. with satisfactory results and it was found to be simple,fast and accurate.
