摘要
[目的]研究转基因拷贝数对转化植株农艺性状的影响。[方法]以农杆菌介导的遗传转化法获得的水稻转基因植株为材料,观察其株高、分蘖、扬花、籽粒形状及饱满度、抗虫性等。同时获取PCR为阳性的单株叶片进行Southern杂交,检测转入基因的拷贝数,研究转基因拷贝数对转化植株农艺性状的影响。[结果]T0代转基因植株的农艺性状有较多改变,如白化、矮化、分蘖少及种子畸型等。转基因植株的转基因拷贝数影响其农艺性状,拷贝数越多农艺性状改变越大,目标基因越容易受到抑制而不表达。在T0代表现感虫的转化植株其T1代也表现感虫。[结论]转基因植物商品化时,应选择单拷贝的转化植株。
[ Objective ] The study was to research the effects of transgene copy number on agronomic characters of transformed plants. [ Method ] With rice transformed plants obtained by genetic transformation method mediated by agrobacterium as materials, their plant height, tillers, anthesis, grain shape and plumpness, insect resistance etc. were observed. At the same time, the leaves per plant whose PCR was positive were taken to conduct the southern hybridization. The copy number of transferred gene was detected and the effects of transgene copy number on agronomic characters of transformed plants were studied. [ Result] The agronomic characters of To generation transgenic plants had much change, such as whitening, dwarfing, low tillering and abnormal seed. The transgene copy number of transgenic plants affected their agronomic characters and the more the copy number was the bigger the change of agronomic characters was, the more easily the target gene was inhibitted not to be expressed. The transformed plants sensitived to insects in To generation were also sensitived to insects in T1 generation. [Conclusion] In commercialization of transgenic plants, the transformed plants with single copy should be selected.
