摘要
按Selye法结扎Wistar大鼠冠状动脉(简称冠脉)左前降支复制早期心肌缺血模型;又经颈外静脉注射垂体后叶素复制冠脉痉挛模型。按心肌不同缺血时间(25min,1h,3h)及对照组将以上各模型均分成4组。每组心肌取心尖及其相邻组织块制成石蜡切片,进行HE染色、链霉菌素一生物素肌红蛋白免疫组织化学染色(LSAB-Mb染色)。结果表明:早期心肌缺血组心肌呈片状肌红蛋白(Mb)缺失;冠脉痉挛组呈多发性、小灶性缺失。所以,LSAB-Mb法可望为冠脉痉挛所致的心肌缺血提供客观形态学证据。
In this study, the model of early myocardial infarction was produced by ligating the left anterior descending part of rat coronary artery and the model of coronary artery spasm (CAS) was produced by external jugular vein injection of vasopressin (VP). Every model mentioned above was divided into three experimental groups according to different time lengths of myocardial ischemia (25 min, 1 hr, 3 hr) and one control group. The apex of myocardium and ad joining slice in each group were used to make paraffin sections, then HE and LSAB-Mb staining are performed. Results show that stretch shape Mb depletion was detected in early myocardial infarction group, and well-definedly focal shape in CAS group. Thus LSAB-Mb method can be expected for use of supplying objective morphologic evidence to myocardial ischemia induced by CAS.
