摘要
目的:探讨T-bet和GATA-3的失衡表达在变应性鼻炎和变应性哮喘中的作用。方法:将30只大鼠随机分成变应性鼻炎组、变应性哮喘组及正常对照组(n=10),相应建立模型。采用逆转录-聚合酶链反应(RT-PCR)方法检测3组大鼠淋巴细胞T-bet和GATA-3 mRNA的表达。结果:变应性鼻炎组和变应性哮喘组大鼠T-bet mRNA的相对表达量分别为(0.12±0.07)及(0.09±0.02),较正常组的(0.60±0.09)减少(P均<0.05);GATA-3 mRNA的相对表达量分别为(1.38±0.15)及(1.41±0.21),较正常组的(0.92±0.21)增多(P均<0.05);而变应性鼻炎组和变应性哮喘组间2个指标比较,差异均无统计学意义(P均>0.05)。结论:T-bet和GATA-3的失衡表达可能是上下呼吸道变应性炎症发生的一个共同因素。纠正其平衡很可能为治疗变应性鼻炎和变应性哮喘提供一条新思路。
Aim: To evaluate the role of imbalanced T-cell-specific transcription factors T-bet and GATA-3 in rats with allergic rhinitis and asthma.Methods:A total of thirty rats were randomly divided into allergic rhinitis(AR) group,asthma(AS) group,and control group(n=10).The expression of T-bet and GATA-3 mRNA in these three groups was detected by reverse transcriptase polymerase chain reaction(RT-PCR).Results: The expression of T-bet mRNA in AR group(0.12+0.07) and AS group(0.09±0.02) was both significantly lower than that of control group(0.60±0.09)(P〈0.05).The expression of GATA-3 mRNA in AR group(1.38±0.15)and AS group(1.41±0.21) was both significantly higher than that of control group(0.92±0.21)(P〈0.05).But there had no significant difference between AS and AR group on T-bet and GATA-3 mRNA expression(P〉0.05).Conclusion: The imbalanced expression of T-bet and GATA-3 may be one of the same factor in upper and lower respiratory inflammation.To retrieve this balance could be a new way in treatment of allergic rhinitis and asthma.
