摘要
为了利用碱蓬在盐激条件下积累脯氨酸的优势,探索新的脯氨酸生产途径,以碱蓬无菌苗的子叶为外植体,利用培养基中不同激素组合和培养条件诱导碱蓬愈伤组织形成,并对野生盐地碱蓬和愈伤组织的脯氨酸含量进行测定。结果表明,盐地碱蓬的叶片在培养基MS+2,4-D 2mg/L+6-BA 1mg/L和光照培养条件下,诱导和继代培养的愈伤组织无论在颜色、质地还是在生长速度上效果最好,并且其愈伤组织的脯氨酸积累也达到最高,大约为野生盐地碱蓬含量的20倍。
In order to use Suaeda salsa 's advantage of proline accumulation under salt stress conditions, explore new ways of proline production, the cotyledon that produced by sterilized seed was used as the explants, then the callus was induced under different medium with plant hormone combinations. At last, proline content of wild Suaeda salsa and callus was determined. The results showed that medium (MS+2,4--D 2mg/L+6--BA lmg/L)which has induced and subcultured callus under lightness has the best effect on either the color, texture or the speed of growth, and its content of proline was more than about 20 times of wild Suaeda salsa.
