摘要
为了观察枸杞子提取液(LBA)对RCS大鼠遗传性视网膜色素变性的治疗作用,将出生后的24只RCS大鼠,随机分为对照组和实验组,每组12只。在实验组RCS大鼠出生后10 d开始喂食1 mg/kg LBA,对照组正常饲养。对照组和实验组RCS大鼠分别在出生后25、35、45 d处死,应用HE染色和TUNEL检测观察视网膜感光细胞的坏死和凋亡,并应用免疫组化的方法检测Caspase-2的表达。结果显示:RCS大鼠实验组25 d和35 d时TUNEL检测阳性细胞数目与对照组相比明显减少(P<0.05);实验组和对照组大鼠生后25 d和50 d时Caspase-2在节细胞层和内核层均有表达,但在25 d时实验组比对照组明显减少(P<0.05)。上述结果提示:在RCS大鼠视网膜变性的早期,LBA通过抑制细胞的凋亡对神经元起保护作用,可以使感光细胞得到更多存留,Caspase-2可促使视网膜变性,LBA则可通过抑制其表达对早期变性的神经元发挥保护作用。
To investigate the neuroprotective effect of the aqueous extract of dried fruits of lycium barbarum (LBA) on the hereditary retinal dystrophy of the Royal College of Surgeon (RCS) rats, a total of 24 postnatal RCS rats were randomly and equally divided into two groups, control group (CG) and experimental group (EG), respectively. EG rats were fed with 1 mg/kg LBA daily 10 days postpartum, and CG rats were fed normally. CG and EG RCS rats were executed at postnatal 25, 35 and 50 days. Then photoreeeptor cells necrosis and apoptosis was assessed by HE staining, and TUNEL detection. The expression of Caspase-2 protein was detected in retina by immunohistochemistry. The results showed that comparing with RCS rats in the CG, 25 and 35 days postpartum, the positive expression of TUNEL in RCS rats in the EG obviously reduced ( P 〈 0.05 ) ; 25 and 50 days postpartum, Caspase-2 positive cells were seen in both ganglion cell layer and inner nuclear layer in both CG and EG rats. The positive expression of Caspase-2 was less seen 25 days postpartum in EG rats than CG rats (P 〈0.05 ). The present results suggest that LBA has neuroprotective effects on the early stage of retinal degeneration of the hereditary retinal dystrophy RCS rats by inhibiting apoptosis, resulting more photoreceptors preserved. Caspase-2 protein may contribute to the retinal degeneration and LBA might protect apoptotic neurons by inhibiting the expression of Caspase-2.
