摘要
药物筛选是现代药物开发流程中测试和获取特定生理活性化合物的一个步骤。毛细管电泳技术由于具有样品消耗量小、速度快、柱效高以及所用溶液体系较接近生物体液组成等特点,已经成为一种非常具有潜力的药物及先导化合物的高效筛选工具。文中就毛细管电泳技术在药物筛选中的最新应用情况进行综述,具体从测定药物解离常数pKa值、药物脂水分配系数(logP)、药物与蛋白质的结合常数的测定以及手性药物筛选等方面进行论述,同时也探讨了微流控芯片技术在高通量药物筛选方面的最新研究进展。
Drug screening is the first step of testing and receiving the compounds with special physiological activities in exploitation flow of modern drugs.Capillary electrophoresis(CE) has become a promising method to highly screen the lead compounds.CE has the features of small sample consumption and fast analysis time,and the buffer used for CE is similar to the body fluid.In this article,the new applications of CE in drug discovery were reviewed from several aspects including dissociation constant pKa,liposolubility constant(logP),the interaction between drugs and proteins,and chiral drug screening.In addition,the development of microfluidics was prospected.
