摘要
用末端转移酶介导的切口末端标记法(TUNEL),检测了大鼠不同器官内的凋亡细胞。结果显示,凋亡细胞在光镜下多以核圆形、深染为特征,呈散在分布。有的凋亡细胞裂解成一个或多个凋亡小体。凋亡细胞数量因器官而异,在细胞增生活跃和细胞更替速度快的器官如小肠、大肠和淋巴结内,凋亡细胞较多,相对稳定的肝、胰等器官凋亡细胞较少。凋亡细胞通常被巨噬细胞或邻近细胞吞噬。该法的优点在于保存组织结构的同时,在原位观察到凋亡细胞,因此可用于细胞学、组织学和病理学的研究,也可作为评估激素或药物对肿瘤疗效的方法。
The TUNEL method was used to detect the apoptotic cells at various tissues of rat,which included stomach,small intestine,large intestine,liver,pancreas,lymph node and so on .Under the light microscope,the apoptotic cells scattering in the tissue were characterized by a round nucleus with dense staining.In some cases the apoptotic cells were splitted into several apoptotic bodies.The number of apoptotic cells varied by the type of tissue,for example,in small intestine,large intestine and lymph node,the number of apoptotic cells was more than that in liver or pancreas,which was closely related to the degree of cell proliferaiton and cell turnover rate in organs.The appoptosis might be found in undifferentiated cells, mature cells and shedding cells from epithelium.All of the apoptotic cells were ended phagocytosis either by macrophages or neighbour cells.The TUNEL method offered certain advantages.It could provide in situ visualization of apoptotic cells at single level while preserving tissue architecture.Moreover,the present technique was demonstrated with reproducibility,specifity and simplicity,which enabled it to be widely used not only in cytology,histology and pathology,but also in evalution of the effects of different homonal and chemothreapeutic drugs in clinic.
