卵巢癌组织谷胱甘肽S转移酶-π表达与化疗耐药的关系 被引量:5

Expression of Glutathione S transferase π in Operative Specimens as Marker of Chemoresistance in Patients with Ovarian Cancer

摘要 目的:探讨卵巢癌组织谷胱甘肽S-转移酶-π(glutathioneS-transferase-π,GST-π)表达和化疗耐药的关系。方法:用免疫组化链霉菌素-生物素技术(labeledstreptavidinbiotinmethod,LSAB)检测了87例卵巢上皮性癌组织和30例正常卵巢上皮组织。本组患者术前均未接受化疗。应用χ2检验和Cox-Mantel模块方法分析卵巢癌组织中GST-π表达与临床病理资料、化疗效应、预后之间的关系。结果:(1)87例卵巢癌组织中,GST-π表达阳性59例,阴性28例,阳性率67.8%。30例正常卵巢上皮组织GST-π表达均呈阴性。(2)卵巢癌GST-π表达与临床病理参数无直接关系。(3)GST-π阳性中有43例(43/59)为化疗耐药,阴性中3例(3/28)为耐药。两组对化疗效应的差异有极显著意义(P<0.005)。(4)卵巢癌GST-π表达阳性患者的生存时间明显比阴性患者的生存时间短(P=0.004)。结论:卵巢上皮性癌组织GST-π表达与化疗耐药明显相关,是预后不良的检测指标。 Objective : To study the relationship between the expression of glutathione S transferase π (GST π) in operative specimens and chemoresistance in patients with ovarian cancer. Methods: The expression of GST π in 87 epithelial ovarian cancer tissues and 30 normal ovarian epithelial tissues was determined with labelled streptavidin biotin method (LSAB). All the patients had not received chemotherapy before operation. We used Chi Square and Cox Mantel test to analyze the relativity between the expression of GST π and clinical pathological data, chemotherapeutic response, prognosis in patients with ovarian cancer. Results:(1) 59 (67 8%) of 87 ovarian cancer tissue were demonstrated to be positive expression with GST π, but all 30 normal ovarian epithelial tissue were negative. (2) There was no direct correlation between the expression of GST π and clinical pathological data. (3) 43(43/59) of GST π positive cases were chemoresistant, while only 3 (3/28) of GST π negative ones were chemoresistant. (4) The difference in the chemotherapeutic response between the two groups was obviously significant ( P <0 005). The survival period of the patients with GST π positive expression was also obviously shorter than that of those with GST π negative expression ( P =0.004). Conclusion : These results strongly suggest that GST π expression in epithelial ovarian cancer tissues is closely related to chemoresistance clinically and it may be served as a useful marker to predict the prognosis of patients.
