利用丙酮/磷酸氢二钾双水相体系分离纯化绿原酸的研究 被引量:12

摘要 利用双水相分配技术,研究绿原酸在普通有机溶剂/无机盐体系中的分配行为。利用紫外分光光度计进行定性定量测量,绿原酸的标准曲线为C=0.67649+21.2607A(R=0.99824),限性范围为51μg/ml<C<255μg/ml。实验采用与水互溶的有机溶剂新型双水相萃取体系即丙酮/K2HPO4·3H2O盐体系研究丙酮加入量与K2HPO4·3H2O加入量对绿原酸分配的影响,确定丙酮/磷酸氢二钾体系萃取绿原酸的最佳条件是:双水相体系的总量为10ml,V丙酮8ml、V水2ml、磷酸盐的质量0.8g。在上述条件下,丙酮/K2HPO4体系的两相分离完全,绿原酸的收率达99.94%。本法溶剂易回收、易处理、操作简便。 The partition behavior of chlorogenic acid in aqueous two-phase systems containing organic solvent and inorganic salt was studied. Ultraviolet spectrophotometer was used for the qualitative and quantitative determination of chlorogenic acid. The standard curve equation is C=0.67649+21.2607A, R^2=0.99824, with the linear range of 51 μg/ml to 55μg/ml. In this study, the aqueous two-phase systems containing acetone and dipotassium hydrogen phosphate was adopted to extract chlorogenic acid. The effects of quantity of acetone and K2HPO4 ·3H2O on the partition coefficient of chlorogenic acid were investigated. It was found that the optimal extraction conditions of chlorogenic acid with the aqueous two-phase systems are as follows: volume of the aqueous two-phase system 10 ml consisting of acetone 8 ml, water 2 ml and dipotassium hydrogen phosphate 0.8 g. The yield of chlorogenic acid is up to 99.94% under the above conditions. This method easy to restoring, dispose and operate.
