竹节参粗多糖的初步纯化工艺研究 被引量:5

摘要 先用乙醇对竹节参粗多糖进行分步醇沉以脱除色素,利用Sevag法、TCA法和木瓜蛋白酶法对其进行脱蛋白,以多糖回收率和蛋白损失率为考察指标,比较三种方法对竹节参粗多糖脱蛋白的效果。实验结果表明,进行5次醇沉,最终乙醇浓度为50%时沉淀出乳白色的粗多糖。Sevag法脱蛋白率80.5%,多糖回收率24%;TCA法脱蛋白率59.8%,多糖回收率47.3%;木瓜蛋白酶法脱蛋白率86.7%,多糖回收率为87.9%。可见,木瓜蛋白酶法能有效去除竹节参粗多糖中的蛋白质。 To study on the removal of pigment and protein in the polysaccharides from Panax japonicus. C. A. Mey, ethyl alcohol was used to strip pigment of polysaccharides. Sevag method, TCA method and caroid method were chosen. Compared with effects of the removed protein and the recovered polysaccharide, the yield of removed protein and the rate of recovered polysaccharides were used to evaluate the factor levels. The results indicated that the milk polysaccharides was acquired by 5 times ethyl alcohol deposit and 50% the ethyl alcohol density, the yield of removed protein and the rate of recovered polysaccharides were 80.5% and 24% in Sevag method,59.8% and 47.3% in TCA method,86.7% and 87.9% in caroid method. The conclusion is that caroid method can effectively remove protein in the Polysaccharides from Panax japonicus. C. A. Mey.
