摘要
[目的]研究重离子和X射线辐照对人舌鳞癌Tb细胞周期影响的规律。[方法]采用X射线和离子束分别辐照人舌鳞癌Tb细胞,X射线照射剂量为0、2、4、6、8Gy;重离子照射剂量为0、0.5、1、2.0、4.0Gy。PI荧光探针标记,流式细胞仪检测不同剂量组受照后在6h、12h、24h的细胞周期变化。[结果]人舌鳞癌Tb细胞在X射线照射后,2.0Gy组激活G1期检测点,而4.0、6.0、8.0Gy组激活G2期检测点。重离子照射后,Tb细胞G2/M期阻滞明显增加,阻滞程度具有剂量和时间依赖性,并且0.5、1Gy组细胞在12~24h时间点出现"崩溃"现象,细胞阻滞解除;2Gy和4Gy组细胞表现为明显的G2/M阻滞,未出现"崩溃"现象。在2Gy辐照时对细胞G2期阻滞率达到70%,相当于6GyX射线辐照。[结论]重离子和X射线对人舌鳞癌细胞周期影响不同,小剂量重离子束具有较高的生物学效应。
[ Objective ] To compare the cell cycle alterations in the human tongue squamous ceil carcinoma Tb in vitro after X ray and carbon ion irradiation. [Methods] Tb cells were irradiated with 12C6+ at dose 0, 0.5, 1, 2.0, 4.0Gy or with X-ray at dose 0. 2, 4, 6, 8Gy. Propidium iodide (PI) staining and flow eytometer (FCM) were used for cell cycle measurement. [ Results] After X-ray irradiation, the Tb cell cycle was arrested in G1 stage for 2 Gy group while 4, 6 and 8 Gy groups were arrested in G2 stage. After ^12C^6+ in'adiation, the Tb cells were inhibited significantly in G2/M, the stagnation increased gradually in a dosage and time dependent manner (P 〈 0. 01 ). The stagnation relieved at 24h for 0.5 Gv. while this phenomenon was not occurred for 2 and 4Gy groups. The rate of stagnation in 2Gy group was 70%. which was equal to 6Gy group by X-ray irradiation. [Conclusion] In comparison with X-ray group, the arrested effects of Tb cells in ^12C^6+ irradiate group are obvious.
