IL-18对肾小管上皮细胞Col-Ⅳ及MCP-1表达和分泌的影响

摘要 目的探讨白细胞介素-18(IL-18)对肾小管上皮细胞Col-Ⅳ及MCP-1表达和分泌影响。方法应用终浓度为0,0.1,1,10,100ng/ml的IL-18处理体外培养人类近端肾小管上皮细胞株(HK-2细胞)24,48,72h,用RT-PCR法检测IL-18对HK-2细胞Ⅳ型胶原(Col-Ⅳ)及单核细胞趋化蛋白-1(MCP-1)mRNA表达;采用ELISA法测定细胞培养上清中Col-Ⅳ及MCP-1分泌水平。结果IL-18呈剂量和时间依赖性地促进HK-2细胞Col-ⅣmRNA和MCP-1mRNA的表达,IL-18呈剂量和时间依赖性地促进HK-2细胞Col-Ⅳ的分泌,呈剂量依赖性地促进HK-2细胞MCP-1分泌,但其蛋白分泌量在24h即达高峰,未见时间依赖关系。结论IL-18可能部分通过促进细胞外基质的过度分泌和炎症细胞趋化,参与促进肾间质纤维化的进程。
