摘要
目的:观察补阳还五汤含药脑脊液对受损皮质神经细胞应激反应的调节作用。方法:在离体培养5天的大鼠皮质神经细胞中分别加入不同的含药脑脊液,37℃孵育24h,然后加入10μmol/L谷氨酸继续孵育60min。通过MTT法检测细胞生存活力;RT-PCR法检测即早基因的表达;扫描电镜观察细胞形态变化。结果:补阳还五汤含药脑脊液能很好的提高细胞生存活力,抑制即早基因的表达。结论:中药复方补阳还五汤能对抗皮质神经细胞的谷氨酸损伤,其机制可能通过抑制即早基因c-fos、c-jun的表达,防止皮质神经细胞凋亡。
Objective: To investigate the protective effect of Buyanghuanwu decoction on damaged cortical neurocyte. Methods: Cerebrospinal fluid with different medicinal content was added into rat's cortical neurocytes, which were then incubated at 37℃ for 24 hours, added 10μmol/L aminoglutaminic acid and incubated for 60 mins. The live activity of neurocytes was detected through MTT, and the expression of immediate early genes were detected through RT-PCR, and cell morphological changes were observed through scanning electron microscope. Results: Buyanghuanwu decoction increased the live activity, inhibited the expression of immediate early gene. Conclusion: Buyanghuanwu decoction has protective effects on cortical neurocyte against injury induced by aminoglutaminic acid, through inhibiting the expression of immediate early gene c-fos, c-jun and preventing the apoptosis.
