TLR4 is Involved in the Apoptosis of Hippocampus Cells in Cerebral Ischemia-reperfusion Mice

摘要 目的探讨Toll样受体4(TLR4)在小鼠脑缺血再灌注损伤中对细胞凋亡的影响。方法昆明小鼠90只,随机分为:假手术组、缺血再灌注组、TLR4抗体阻断组,每组依据缺血后再灌注6h、12h、24h、48h、72h不同时间点再分五个小组。采用免疫组织化学方法,Western blot方法检测Caspase-3在各组海马不同再灌注时间点的表达变化,应用TUNEL法检测海马细胞凋亡情况。结果缺血再灌注组Caspase-3蛋白表达量和凋亡细胞数明显高于假手术组(p<0.01),而TLR4阻断组Caspase-3蛋白表达量和凋亡细胞数则明显少于缺血再灌注组(p<0.01)。结论阻断TLR4可减少脑缺血再灌注损伤中的细胞凋亡。 Objective To study the effect of Toll-like receptor 4(TLR4) on the apoptosis in cerebral ischemiareperfusion(I/R) mice. Methods 90 kun-ming mice were randomly divided into sham group, ischemia-reperfusion group and TLR4-bloeked group in different time points of reperfusion (6h,12h, 24h, 48h and 72h). The expression of caspase-3 was studied by immunohistochemistry and Western blot, the apoptotic hippocampus ceils were stained by TUNEL. Results Both the expression of caspase-3 and the number of apoptosis cells increased significantly in I/R group than in sham group(p〈 0.01), however the expression of easpase-3 and the number of apoptosis cells decreased in TLR4-blocked group compared with I/R group (p〈 0.01). Conclusion Blocking TLR-4 can decrease the cells apoptosis caused by ischemia-reperfusion.
