摘要
目的观察骨髓源性干细胞移植对慢性马兜铃酸肾病(CAAN)大鼠肾小管功能损害的干预作用。方法制作慢性马兜铃酸肾病大鼠模型,随机分为治疗组和非治疗组,治疗组予以经尾静脉注射同系雄性大鼠骨髓干细胞;非治疗组予以尾静脉注射等量生理盐水;另设正常对照组。各组每周测量体重,并分别于尾静脉注射干细胞或生理盐水后第30d留尿,测24h尿量、用酶-底物直接显色法检测尿N-乙酰-β-D-氨基葡萄糖苷酶(NAG)量和用放射免疫法检测尿微球蛋白(β2-MG)。结果治疗组尿NAG含量及尿β2-MG排泄量较非治疗组明显减少,差异显著(P<0.05),但仍高于正常组(P<0.05)。骨髓干细胞移植能减少慢性马兜铃酸肾病大鼠尿NAG酶和尿β2-MG的排泄量。结论骨髓干细胞移植对慢性马兜铃酸肾病肾小管功能损害有一定的恢复作用。
Objective To study the effect of bone marrow derived stem cells(BMDSCs) transplantation on the renal tubules function in the rats of chronic aristolochic acid nephropathy(CAAN). Methods To make the CAAN animal model of female Wistar rats, then the CAAN rats were divided into treated and untreated groups in random. The rats of treated group were transplanted with BMDSCs of the homologous series male rats through the caudal vein. The rats of untreated group were intravenously injected with Sodium Chloride through the caudal vein, and of nomal control group treated with nothing. The rats' body weights were measured weekly, their 24h-urine was collected 30 days after injection, the contents of urine β2 - MG and urine NAG were determined with radioimmunity and enzyme-substrate respectively. Results The levels of urine 2 - Mg and NAG were decreased in the treated rats compared with the untreated rats(P〈0.05), but still higher than those in nomal rats(P〈0.05). The transplantation of BMDSCs can decrease the contents of urine β2 - MG and urine NAG in the CAAN. Conclusion BMDSCs have recovery effect on the renal tubules of the chronic aristoloehie acid nephropathy.
