摘要
目的:探讨采用超滤法分离技术纯化葫芦巴中葫芦巴总皂苷的工艺。方法:以浸膏得率和薯蓣皂苷元的含量为评价指标进行正交实验设计,对操作压力、滤膜的分子截流值、药液浓度、药液温度4个因素进行优选。结果:选择的最佳工艺为A_3B_2C_3D_2,即操作压力为2kg·m^(-2),膜孔径(以截流分子量计)10 000,药液浓度为生药量1.0g·mL^(-1),药液温度为25℃时纯化效果最好。结论:所选择的膜分离技术纯化葫芦巴中葫芦巴总皂苷的工艺操作简单可靠,纯化效率高。
Objective: To isolate and purify Saponin from Trigonella foenum-graecum by uhrafihration. Methods: orthogonal experiment was designed by taked extract rate and diosgenin content as the evaluating indicator and used four factors to optimize which were operation of pressure ( A ), average membrane flux ( B ), concentration of chemical(C) and (D). Results: The optimum technology is A3 B2 C3 D3, namely, operation of pressure 2.0 kg· 4^-2 , iris aperture 10000(stop-flow molecular) , concentration of chemical 1.0g·mL^-1 ( crude drug) , temperature of chemical 25℃. Conclusion: The technology is simple and reliable and have higher purification efficiency.
