双向电泳技术研究镉诱导后牙鲆肝的差异蛋白质组

Differential Proteome of Liver in Paralichthys olivaceus Induced with Cadmium Revealed by Two-dimensional Gel Electrophoresis

摘要 采用改良的TCA/丙酮法直接提取牙鲆肝脏蛋白质组,并采用双向凝胶电泳技术予以分离.实验结果表明:经氯化镉诱导后,牙鲆肝表达出16个差异蛋白质(斑点),其中4个斑点上调,2个斑点下调,6个斑点消失,新增4个斑点;并且蛋白质斑点在凝胶上的分布表现出较高的重复性.一旦这些差异蛋白质点在双向凝胶电泳图谱上实现标准指纹化后,其图谱中的差异蛋白斑点分布规律可用于监测与评价流动水体中镉污染程度及危害性. The liver proteome in Paralichthys olivaceus (PO) was directly extracted by a modified TCA/acetone method, and separated by two-dimensional gel electrophoresis (2-DE). The results showed that the liver of PO expressed 16 differential proteins (protein spots) after cadmium chloride inducement, 4 spots of which were up-regulated, 2 down-regulated, 6 repressed and 2 induced. In addition, we found that these differential spots and their orientation showed repeated results in the 2-DE gel. Once these spots of differential proteins in the 2D- PAGE gel were located by standard fingerprint technique, the distribution regulation of these spots in gel map could be directly used to monitor as well as evaluate the contamination level of cadmium and its critical level in halobios.
