摘要
目的观察苦参碱对人急性髓白血病细胞株KG1a体外增殖抑制和凋亡的影响。方法CCK-8法和台盼蓝拒染色法检测苦参碱对KG1a细胞增殖抑制作用和细胞存活率;甲基纤维素培养体系分析细胞克隆形成能力;流式细胞仪法检测作用前后KG1a细胞的凋亡率及细胞周期变化。结果苦参碱能够抑制KG1a细胞增殖,随着药物浓度增大和作用时间延长,抑制率逐渐升高、存活率逐渐下降;作用后细胞克隆形成率明显低于对照组(P<0.05);各浓度药物组均能诱导KG1a细胞凋亡;苦参碱作用后细胞大部分被阻于G0/G1期。结论苦参碱能抑制KG1a细胞的增殖,改变其周期,诱导其凋亡。
Objective To observe the effects of Matrine on anti-proliferation and apoptosis of leukemia cell lines KGla. Methods The growth inhibiting and rate of cell survival were measured by CCK-8 assay and trypan blue exclusion test. The clonality was assayed in culture of cellosolve system. The rate of apoptosis and cell cycle of KG1 a cell were assayed by flow eytometry before and after treated by Matrine. Results Matrine could inhibit KG1 a cell growth, with the increase of drug concentration and time extension, the inhibition rate gradually increased and survival rate gradually decreased. The elonality was dramatically lower than that in control group (P 〈 0.05 ). The concentration of drug group could induce apoptosis KGla cells. KGla cell was arrested at G0/G1 stage treated by Matrine. Conclusion Matrine can inhibit cell proliferation of KGla, change its cycle and induce its apoptosis.
