摘要
目的:研究药物代谢酶基因多态性对丙戊酸钠代谢的影响,并建立分析丙戊酸代谢酶中CYP2C9、UGT1A6的基因多态性的方法。方法:从人全血中抽取gDNA,设计引物扩增CYP2C9及UGT1A6,并运用限制性片段长度多态性(RFLP)技术的方法进行CYP2C9*2、CYP2C9*3、CYP2C9*4、CYP2C9*5、UGT1A6*2和UGT1A6*3的分型。结果:成功设计了引物用来扩增包含CYP2C9*3、CYP2C9*4、CYP2C9*5的片断,并测序验证。结论:建立了简单可靠的RFLP基因分型方法。
Objective: In order to study the effect of drug-metabolizing enzymes (DME) polymorphism on the metabolism of valproie acid ( VPA), we set up a method to analyze the DME gene polymorphism. Methods:The GDNA were extracted from blood of human. The CYP2C9 and UGTIA6 gene were amplified with PCR. We used the restriction fragment length polymorphism (RFLP) to classify the CYP2C9 * 2, CYP2C9 * 3, CYP2C9 *4, CYP2C9 * 5, UGTIA6* 2 and UGT1A6 *3 genotypes. Results:A successfttl design of the primers was used to contain expansion of CYP2C9* 3, CYP2C9 * 4, CYP2C9 * 5 of the dips, and sequencing and verification were performed. Conclusions:We established a simple and reliable RFLP method to group the UGT1A6 and CYP2C9.
