基质金属蛋白酶9和组织金属蛋白酶抑制剂1在大鼠肾脏机械性损伤后的表达

摘要 目的探讨大鼠肾脏机械性损伤后肾组织基质金属蛋白酶9(MMP-9)和组织金属蛋白酶抑制剂1(TIMP-1)的表达及意义。方法36只Wistar大鼠随机分为对照组和创伤组。采用自由落体生物撞击仪撞击大鼠脊肋角复制创伤动物模型。用组织芯片技术及免疫组织化学方法检测各组大鼠创伤后1、6、12、24和36h肾组织中MMP-9和TIMP-1蛋白的表达。结果对照组肾组织中MMP-9蛋白表达微弱,创伤组MMP-9表达在创伤后1h开始增强,6h迅速增加并达高峰(P<0.05),以后逐渐下降并接近正常。创伤组各时段TIMP-1蛋白表达与对照组相比,差别无统计学意义(P>0.05)。结论大鼠肾脏机械性损伤后MMP-9是参与肾脏损伤的重要因素之一。 Objective To investigate the expressions and significance of matrix metalloprotease-9 (MMP-9) and tissue inhibitor of metalloproteinase-1 (TIMP-1) in kidneys of rats after mechanical injury. Methods Thirty-six adult female Wistar rats were randomly divided into two groups: 6 in control group and 30 in trauma group. The model of trauma rats was constructed by striking the spinal column and ribs of rats with free-falling bio-impactor. MMP-9 and TIMP-I expressions in renal tissues of each group were detected by tissue chips and immunohistochemistry at 1, 6, 12, 24 and 86 h after trauma, Results MMP-9 group while it began to increase at 1 h, reached expression of renal tissues was lower in control high peak at 6 h (P 〈0.05) and then dropped gradually to normal in trauma group. TIMP-1 remained relatively unaltered in trauma group Conclusion MMP-9 may play an important role in kidney injury after mechanical injury.
