血红素加氧酶-1和热休克蛋白70在肾移植中的表达

摘要 目的探讨血红素加氧酶-1(HO-1)和热休克蛋白70(HSP70)在大鼠肾移植急性排斥反应中的表达及意义。方法建立大鼠同种异体肾移植模型。实验分为Ⅰ组(正常对照)、Ⅱ组(同基因移植:SD大鼠→SD大鼠)、Ⅲ组(异基因移植:Wistar大鼠→SD大鼠)、Ⅳ组(异基因移植环孢霉素A干预)。用免疫组化法检测移植肾组织中HO-1和HSP70的表达,并观察其与病理学检查的相关性。结果同基因移植组移植肾中HO-1和HSP70的表达水平在术后无明显变化(P>0.05);异基因移植组移植肾中HO-1和HSP70的表达在术后第1、3天和第7天逐渐升高(P<0.01),且与病理分级间存在正相关(r=0.911,P<0.01;r=0.869,P<0.01)。结论HO-1和HSP70参与肾移植急性排斥反应,监测HO- 1和HSP70的表达可能为急性排斥反应的早期诊断和治疗提供新的方法。 Objective To investigate the expressions of hemeoxygenase-1 (HO-1) and heat shock protein 70 (HSPT0) in renal allograft of rats. Methods The rat model of allogene renal transplantation was established. In four groups, normal control group, homogenic transplantation group (SD→SD), allogene transplantation group (Wistar→SD) and CsA group, the expression of HO-1 and HSP70 in kidney was detected by immunohistochemistry, the correlation between HO- 1 as well as HSP70 and pathological findings was observed. Results The expression levels of HO-1 and HSP70 in the homogenic transplantation group did not change greatly (P 〉0.05) but increased gradually on day 1, day 3 and day 7 in the allogene transplantation group (P 〈0.01), and there were significant correlation between the two factors and pathological indexes in the allogene transplantation group (r=0.911, P 〈0.01; r=0.869, P 〈0.01). Conclusion HO-1 and HSP70 may play important roles in acute renal allograft rejection. Detection of HO-1 and HSP70 expressions in allograft may be useful for early diagnosis and treatment of acute rejection following renal transplantation.
