摘要
3-脱氧葡糖醛酮(3-Deoxyglucosone,3-DG)是一种高反应性的毒性2-羰基醛化合物,在3-DG还原酶作用下可脱毒生成3-脱氧果糖,在羰基醛脱氢酶作用下脱毒生成3-脱氧葡萄糖醛酸。3-DG代谢酶系统作为一种自身防御系统,可能通过调节3-DG而减少美拉德反应的进行和晚期糖基化终末产物(Advanced glycation end products,AGEs)过量生成,并使3-DG处于相对正常水平,这对防止老化病、糖尿病、尿毒症等方面均有积极意义。本文对3-DG代谢酶的来源、分布情况,酶的种类、部分酶学性质及其与临床疾病的关系等研究进展做简要综述。
3-deoxyglucosone(3-DG) ,a highly reactive and cytotoxic 2-oxoaldehyde compound,is detoxified mainly to 3-deoxyfructose by 3-DG- reducing enzymes, or to 2-keto-3-deoxygluconic acid by oxoaldehyde dehydrogenase. 3-DG metabolizing enzymes, self-defence enzymes, may keep the concentrations of 3-DG at normal level by adjusting the reactive 2-oxoaldehyde compounds like 3-DG,preventing the advanced stage of the Maillard reaction and inhibiting the overproduction of advanced glycation end products (AGEs). These evidences suggest that 3-DG metabolizing enzymes have played an active role in preventing the development of aging,diabetes and uremia. This paper reviews the sources ,distribution,kinds,enzymatic character of 3-DG metabolizing enzymes and their relationship with clinical diseases associated with 3-DG.
