补骨脂等5种中药植物雌激素活性的实验研究 被引量:34

Phytoestrogenic activity of five kinds of Chinese herbal medicines including Fructus Psoraleae

摘要 目的评价补骨脂等5种中药的植物雌激素活性及其可能机制。方法正常Wistar雌性大鼠(刚断乳)96只,随机分为12组(溶剂对照组、己烯雌酚组、5个中药组和5个中药+己烯雌酚组)。各组动物给药4d后处死,计算动物子宫系数和体重增幅;取血并分离血清。利用雌激素受体(ER)阳性MCF7细胞,以MTT法检测含药血清对细胞增殖的影响;通过流式细胞术检测含药血清对细胞周期的影响。结果①灌胃补骨脂、川牛膝能明显增加大鼠的子宫系数(P<0.05);灌胃补骨脂、川牛膝、红花可使己烯雌酚引起的大鼠子宫系数的升高受到显著抑制(P<0.05);灌胃川牛膝可使大鼠的体重增幅明显上升(P<0.05);②补骨脂、川牛膝、红花、丹参等中药组含药血清均可使MCF7细胞增殖加速(P<0.01),加入ERβ激动剂DPN可减弱其促增殖效应;在拮抗实验中,中药组含药血清对细胞增殖的促进作用较单独使用己烯雌酚的组别有明显下降,加入DPN可进一步增强其下降效应(P<0.05或P<0.01);③补骨脂、川牛膝、丹参、菟丝子等中药组含药血清均可提高细胞增殖指数;各中药+己烯雌酚组含药血清对己烯雌酚引起的细胞增殖指数的增加均无明显的拮抗作用。结论补骨脂、川牛膝、红花、丹参、菟丝子等5种中药均具有一定的植物雌激素作用,但其作用强弱及途径有所差异。 Objective To review the (CHM) including Fructus Psoraleae phytoestrogenic activity of five kinds and possible mechanism. Methods of Chinese herbal medicines All 96 normal Wistar female rats (just weaning) were randomly divided into 12 groups (solvent control group, diethylstilbestrol group, 5 groups of CHM and 5 groups of CHM combined with diethylstilbestrol). After 4 days all rats were killed for taking blood to extract the serum samples. The rat uterus coefficients and the gain in weight were calculated. The influence of the medical serum on cell proliferation was detected by using MTT assay in estrogen receptor (ER) positive MCF7 cells, and the effect of it on cell cycle was tested by using flow cytometry. Results O)Fructus Psoraleae and Radix Cyathulae administrated intragastrically increased significantly the rat uterus coefficient ( P 〈 0.05 ), while Fructus Psoraleae, Radix Cyathulae and Flos Cartharni administrated intragastrically inhibited significantly the increase of rat uterus coefficient induced by diethylstilbestrol ( P 〈 0.05 ). Radix Cyathulae administrated intragastrically raisedrat weight significantly ( P 〈 0.05 ). ②In the groups of CHM the medical serum containing Fructus Psoraleae, Radix Cyathulae, Flos Carthami or Radix Salviae Miltiorrhizae promoted the cell proliferation of MCF7 ceils (P 〈 0.01 ), but promotion effect could be reduced by ERβ sagonist DPN. In the antagonism test the promotion effect of medical serum in the groups of CHM decreased significantly than that in the diethylstilbestrol group, and DPN could improve this decreasing effect furthermore (P 〈 0.05 or P 〈 0.01 ). ③The medical serum containing Fructus Psoraleae, Radix Cyathulae, Radix Salviae Miltiorrhizae or Semen Cuscutae increased the indexes of cell proliferation in the groups of CHM. The medical serum had no obvious antagonism effect on the increase of cell proliferation indexes induced by diethylstilbestrol in the groups of CHM combined with diethylstilbestrol. Conclusion Fructus Psoraleae, Radix Cyathulae, Flos Carthami, Radix Salviae Miltiorrhizae or Semen Cuscutae have phytoestrogen-like effects, but they are different in intensity and action pathway.
