摘要
目的观察溃疡平对胃溃疡大鼠胃黏膜组织表皮生长因子(EGF)和表皮生长因子受体(EG-FR)mRNA表达的影响,探讨溃疡平的促进胃溃疡愈合的机制。方法采用改良Okabe法制备慢性乙酸胃溃疡模型;随机分为:实验对照组,模型组,溃疡平高、中、低剂量组,盐酸雷尼替丁组,共6组;给药治疗14 d,采用实时荧光定量PCR法检测胃黏膜组织EGF和EGFR mRNA表达。结果与模型组相比,溃疡平低、中、高剂量组的EGF 2-ΔΔCT值明显增高,表明溃疡平能使EGF mRNA表达上调,有统计学意义(P<0.01);溃疡平低、中、高剂量组的EGFR 2-ΔΔCT值明显增高,表明溃疡平能使EGFR mRNA表达上调,有统计学意义(P<0.001)。结论溃疡平可促进胃溃疡大鼠胃黏膜组织EGF和EGFR mRNA表达,这可能是其促进溃疡愈合,提高溃疡愈合质量,减少溃疡复发的分子学基础之一。
Objective To investigate the curative mechanism of Kuiyangping by studying the effect of iton expressions of EGF and EGFR mRNA in gastric mucosa in rats with experimental gastric ulcer(GU). Methods The ulcer model was prepared by modified Okabe's method. The rats were randomly divided into 6 groups : experimental group, the model group, the low, middle and high dose of Kuiyangpin groups and the ranitidine hydrochloride group. After the 14-day treatment, the expressions of EGF and EGFR mRNA in the mucosa were measured by real-time PCR. Result Compared with the model group, the value of EGF 2^-ΔΔCTT in the low, middle and high dose of Kuiyangping groups were obviously enhanced, which indicated that Kuiyangping could up-regulate the expression of EGF(P 〈0.01 ) ; and the value of EGFR 2^-ΔΔCT in the low, middle and high dose of Kuiyangping groups were obviously enhanced, which indicated Kuiyangpin could up-regulate the expression of EGFR(P 〈 0. 001 ). Conclusion Kuiyangping can increase the expressions of EGF and EGFR mRNA in gastric mucosa in rats with gastric ulcer, which may be one of the molecular basises of its functions to accelerate ulcer healing, improve the quality of healing and reduce ulcer recurrence.
