电针对佐剂性关节炎大鼠炎症局部阿片肽基因表达的影响 被引量:18

Effects of Electro-acupuncture on Opioid Peptides Gene Expression of Inflamed Location in Adjuvant Arthritic Rats

摘要 以佐剂性关节炎大鼠为炎症痛模型,选择悬钟、昆仑穴位,采用原位杂交组织化学方法,观察电针(EA)对大鼠炎症局部阿片肽前体物质前阿黑皮素(POMC)和前脑啡肽原(PENK)mRNA的表达.结果表明EA可以促进炎症局部免疫细胞中POMC和PENKmRNA的表达,使阿片肽的合成和释放增加,以实现外周炎症局部的镇痛作用. Previous studies suggested that endogenous opioid peptides (EOP) participate in the peripheral mechanism of electro-acupuncture (EA) analgesia for inflammatory-pain.This study observed the effects of EA on EOP gene expression of inflamed location in inflammatory-pain rats,further sought for peripheral mechanism of EA analgesia.The experiments used adjuvant arthritic (AA) rats as the inflammatory-pain model,selected Xuanzhong and Kunlun points,adopted detective methods of in situ hybridization histochemistry to observe mRNA expression of the EOP precursor,pro-opiomelanocortin (POMC) and proenkephalin (PENK),in inflamed location and etc.The results showed that EA promotes gene expression of the EOP precursor while analgesic,accelerates releases of EOP.The results also provide some theoretical basis for the effect accumulation of acupuncture analgesia,and have certain reference value for peripheral analgesic mechanisms of EA in chronic pain.
