环氧树脂包埋块半薄切片进行免疫组化染色的方法 被引量:2

摘要 为应用环氧树脂包埋的电镜制样切片进行光镜免疫组化观察,探讨脱树脂试剂的浓度、新鲜度、作用时间及切片的厚度对Epone-812环氧树脂包埋的人体子宫平滑肌组织经氧化醇钠脱树脂处理后,进行ABC法α-肌动球蛋白免疫组化染色效果的影响。结果显示新鲜配制的过饱和氧化醇钠,作用5min其脱树脂效果良好且不掉片,免疫组化染色显色明显、清晰。表明脱树脂试剂的浓度、新鲜度和作用时间是实验成功的关键因素。切片厚度未见对免疫组化染色有明显影响。 In order to us the semithin sections embedded with epoxy resin for immunolabelling, the deresin conditions including concentration, freshness and treatment duration of sodium ethoxide, an epoxy resin remover, were studied. The ABC method of immunohistochemical staining was used to detect the exist of α-actin on the semithin sections of human uterine smooth muscles embedded with Epone-812. The effects of three sodium ethoxide preparations were compared. Only 5min of treatment with oversaturated and freshly prepared sodium ethoxide removed the embedding medium completely and optimal immunolabeling was achieved. The result indicated that the concentration, freshness and treatment duration of deresin reagent were the key factors for the experiment. The thinness of the sections did not affect the immunohistochemical staining.
