摘要
采用PCR-SSCP技术分析了MyoD基因在3个西南地方黄牛品种的遗传多态性。结果表明:MyoD的第2外显子内不存在遗传多态性;第1外显子在3个牛品种中检测到了AA、AB和BB基因型,而且A等位基因为3个牛群体的优势等位基因,分布较高。3个品种中,盘江牛AA基因型频率最高,达到0.5365,而巴山牛和昭通牛则相对较低,分别为0.4465和0.2564。对第1外显子的多态片段测序分析表明:位于MyoD基因第782bp处发生碱基G→A的突变,并导致了氨基酸的突变,使甘氨酸变为丝氨酸。
The polymorphism of the MyoD gene in Panjiang cattle, Zhaotong cattle and Bashan cattle were analyzed by PCR-SSCP. The results showed that there were no polymorphism detected by the exon 2 of the bovine MyoD and there were three genotypes (AA,AB and BB) detected by the exon 1 of the bovine MyoD in three breeds. The frequency of A allele was higher than that of B allele in three breeds. The frequency of AA genotype was the highest in three breeds, which Panjiang cattle was 0.5294. The Bashan cattle and Zhaotong catle were 0.4465 and 0.2564, respectively. The polymorphism fragments amplified were sequenced. The sequencing results indicated that there was one single nucleotide mutation G→A at 782 bp of MyoD gent. And this mutation resulted in amino acid Gly Changing into Ser.
