摘要
目的:研究10-羟基喜树碱注射液在小鼠体内的药动学特征。方法:建立测定小鼠血浆中10-羟基喜树碱内酯形式的HPLC-荧光检测法.小鼠尾静脉注射和腹腔注射后不同时间点进行眼眶静脉丛取血,测定血浆中内酯形式的血药浓度,并用PKBP-N1药动学程序对血药浓度进行处理。结果:10-羟基喜树碱内酯形式可与血浆中的其它成分较好地分离,在1.25~2500ng·ml^(-1)的血药浓度范围内呈良好的线性关系,10-羟基喜树碱两种给药方式在小鼠体内的药动学特征基本相似。结论:与静脉注射给药方式比较,腹腔注射可以提高局部组织的药物浓度.减轻药物对全身的副作用。
Objective: To study the pharmacokinetic characteristics of, 10- hydroxycamptothecin(HCPT) injections in rats.Methods: A reversed-phase high-performance liquid chromatography with fluorescence detection was developed and validated for analysis of HCPT in lactone in rat plasma. The plasma concentrations of HCPT in lactone was detected at various times roll,ring a single iv and a single ip dose of 6mg/kg HCPT. The data were processed with the pharmacokinetic software PKBP- N1 .Results: A good separation of HCPT in lactone forms with other substances in plasma was obtained. The assay was liner over the range of 1.25 - 2500ng·ml^-1. There was no significant difference of pharrnacokinetic characteristics in rats between two ways. Conclusion: Compared with iv , ip can greatly increase drug concentration in tissues, decrease side effects.
