P21 WAF1/CIP1 在大肠癌中的表达

P21 WAF1/CIP1 Expression in Colorectal Carcinoma

探讨Ｐ２１ＷＡＦ１／ＣＩＰ１与ｐ５３基因在大肠癌中的表达意义及其关系。方法应用原位杂交及免疫组化技术检测大肠癌中的抑癌基因Ｐ２１ＷＡＦ１／ＣＩＰ１ｍＲＮＡ及ｐ５３蛋白的表达。统计学采用卡方检验。结果发现大肠癌组织及癌旁组织中Ｐ２１ＷＡＦ１／ＣＩＰ１表达阳性率分别为８４．１３％（５３／６３）及６１．９０％（３９／６３），两者之间差别显著（Ｐ＜０．０５）。Ｐ２１ＷＡＦ１／ＣＩＰ１表达程度与癌组织的分化程度有关，两者之间存在显著差别（Ｐ＜０．０５）。Ｐ２１ＷＡＦ１／ＣＩＰ１表达与其他临床病理指标无关（Ｐ＞０．０５）。ｐ５３蛋白在大肠癌中阳性率为３８．０９％（２４／６３），ｐ５３过表达的患者其Ｐ２１ＷＡＦ１／ＣＩＰ１的表达较之ｐ５３阴性者为低，二者存在显著差异（Ｐ＜０．０５）。结论Ｐ２１ＷＡＦ１／ＣＩＰ１作为一新的抑癌基因，其表达相当程度上依赖于ｐ５３蛋白。

Objective\ To investigate the expression of P21 WAF1/CIP1 gene and its relationship to the p53 gene expression.\ Methods\ Hybridization in situ and immunohischemical techniques were used.\ Results\ The positive rate of P21 WAF1/CIP1 expression in colorectal carcinoma and normal mucosa was 84.13% and 61.9% respectively,and was significantly different(P<0.05). The P21 WAF1/CIP1 expression intensity reflected to the tumor differenciation,but not to the depth of tumor invasion,lymph node metastasis and pathological stage.\ The p53 positive rate was 38.09%.\ The P21 WAF1/CIP1 expression rate in the p53 positive group was lower than that of the p53 negative group(P<0.05).\ Conclusion\ The P21 WAF1/CIP1 is a suppression gene of tumor,its expression depends on the p53 largely.
