肉毒碱复合高脂饲料喂养兔血脂水平的变化 被引量:1

摘要 背景:肉毒碱是脂肪酸进入线粒体内进行氧化代谢的惟一转运载体,其在机体内水平对血脂代谢具有重要的影响作用。目的:观察肉毒碱对高脂饲料喂养兔血脂水平变化的影响。设计、时间及地点:随机对照动物实验,于2006-10/2007-05在解放军南京军区南京总医院检验科及比较医学科完成。材料:普通级健康纯系雄性新西兰兔14只,体质量(1.98±0.22)kg,用于建立兔高脂血症模型。方法:14只雄性新西兰兔随机抽签法分为对照组及实验组,每组7只。对照组仅用高脂饲料喂养,实验组用高脂饲料喂养的同时每天补充肉毒碱200mg/kg体质量。分别于实验开始后第0,2,3,4周称体质量并抽取耳动脉血,分离血清后测定血中总胆固醇、三酰甘油、高密度脂蛋白胆固醇、低密度脂蛋白胆固醇以及载脂蛋白B和血清游离肉毒碱水平。主要观察指标:不同时间点各组动物体质量的变化以及血中总胆固醇、三酰甘油、高密度脂蛋白胆固醇、低密度脂蛋白胆固醇以及载脂蛋白B和血清游离肉毒碱水平。结果:14只新西兰兔全部进入结果分析。4周内,实验组和对照组体质量差异无显著性意义(P>0.05)。自第2周开始,实验组兔血清游离肉毒碱水平高于对照组(P<0.01);总胆固醇、载脂蛋白B水平明显低于对照组(P<0.05或P<0.01);3周后,实验组血清三酰甘油、低密度脂蛋白胆固醇水平也低于对照组(P<0.01);两组高密度脂蛋白胆固醇水平差异无显著性意义(P>0.05)。结论:补充服用肉毒碱能够抑制高脂饮食导致的血中总胆固醇、三酰甘油、低密度脂蛋白胆固醇以及载脂蛋白B水平升高。 BACKGROUND: Carnitine, which is the only carder for the fatty acid crossing into mitochondria for oxidize, has important effect on serum lipids metabolism. OBJECTIVE: To investigate the effect of carnitine supplement on the level changes of serum lipids in rabbits fed with high-fat diet. DESIGN, TIME AND SETTING: The randomized controlled animal experiment was performed at the Department of Clinical Laboratory, General Hospital of Nanjing Military Area Command of Chinese PLA from October 2006 to May 2007. MATERIALS: 14 male New Zealand rabbits, weighting (1.98±0.22) kg, were adopted to establish hyperlipidemia model. METHODS: 14 rabbits were randomly divided into experimental and control group, with 7 animals in each. Rabbits in the control group were fed with high-fat diet, and those in the experimental group were supplied with 200 mg/kg carnitine everyday combined with high-fat diet. The blood was collected from central artery and body weight was obtained at 0, 2, 3, 4 week after the experiment, then total cholesterol, triglyceride, high density lipoprotein cholesterol, low density lipoprotein cholesterol, apolipoprotein B and free carnitine levels in serum were measured. MAIN OUTCOME MEASURES: Changes of body wight, total cholesterol, triglyceride, high density lipoprotein cholesterol, low density lipoprotein cholesterol, apolipoprotein B and free carnitine levels in serum were measured at various time points in each group. RESULTS: All 14 rabbits were involved in the result analysis. Body weight of the rabbits between the experimental group and control group showed no difference in 4 weeks (P 〉 0.05). From the second week, serum carnitine level in the experiment group was higher than that of the control group (P 〈 0.01), total cholesterol and apolipoprotein B levels were significant lower than those in the control group (P 〈 0.05 or P 〈 0.01). After the third week, the levels of triglyceride and low density lipoprotein cholesterol in the experiment group were lower than those in the control group (P 〈 0.01). There was no significant difference of high density lipoprotein cholesterol level between the two groups (P 〉 0.05). CONCLUSION: Carnitine supplement can inhibit the increasing of total cholesterol, triglyceride, low density lipoprotein cholesterol and apolipoprotein B levels in serum of the rabbits fed with a high-fat diet.
