摘要
目的:应用原子力显微镜(AFM)和透射电镜观察低强度电磁场(LIPEMF)对表皮干细胞超微结构的影响。方法:对经LIPEMF作用5d(选取频率50Hz,5mT,30min/次,1次/d)和未经LIPEMF作用的体外培养的人表皮干细胞,用AFM和透射电镜进行细胞超微结构观察。应用AFM,两组分别测量10个表皮干细胞的细胞直径、细胞表面平均粗糙度和均方根粗糙度。结果:从AFM可见,人表皮干细胞表面分布着大量直径几十到几百纳米不等的小颗粒,经LIPEMF作用的表皮干细胞表面的颗粒高度、粗糙度等较未经LIPEMF作用的细胞高;透射电镜发现作用组细胞稍较成熟,核浆比例小,胞质细胞器稍多。结论:低强度电磁场对人表皮干细胞的超微结构有一定影响,AFM有助于观察其表面超微结构变化。
Objectives:To study the influence of the low intensive pulse magnetic field (LIPEMF) on ultrastructure of epidermal stem cells (ESCs). Methods: The morphologic features and ultrastmcture of ESCs stimulated by LIPEMF (50Hz, 5mT, 20min, 1 time/d) for 5 days and those untreated were observed under AFM and transmission electron microcopy. Cell diameter, mean square roughness and average roughness of cell membrane between two groups were measured and compared. Results:Under AFM, many particles,with different diameters distributed in ESCs. The mean square roughness and average roughness of ESCs membrane were more heavy in LPEMF group than that in control group. Transmission electron microcopy showed that ESCs stimulated by LPEMF were more mature, with small proportion of karyon to cytoplasm, and more organelles in cytoplasm, compared to that of control. Conclusions: The low intensive pulse magnetic field effects on ultrastructures and morphological features of ESCs, which can be well studied by AFM.
