摘要
目的:探讨大肠癌组织中Survivin蛋白和CD34表达及其意义。方法:应用免疫组织化学方法检测正常大肠粘膜、腺瘤和腺癌组织中Survivin和CD34表达,计数CD34标记的微血管密度(MVD)。结果:Sur-vivin蛋白在正常大肠粘膜无表达,在腺瘤和腺癌组织中表达的阳性率分别为62.5和70.9,大肠腺瘤组和大肠癌组中Survivin表达率显著高于正常大肠粘膜组;大肠腺瘤和腺癌组织中MVD计数分别为21.55±10.61和28.89±10.47,显著高于正常大肠粘膜组的8.23±2.77,三者之间有显著性差异;Survivin蛋白和CD34表达与肿瘤浸润深度和淋巴结转移明显相关;Survivin蛋白表达与MVD呈正相关。结论:Survivin蛋白可能通过抑制细胞凋亡,诱导新生血管形成而影响大肠癌的发生和发展,Survivin蛋白和CD34表达可作为判断大肠癌预后的指标。
Objective:To investigate the expression and significance of Survivin protein and CD34 in colorectal carcinoma. Methods :The expression of Survivin and CD34 in normal mucosa, colorectal adenoma and adenocarcinoma were detected by immunohistochemical method. Microvessel density(MVD) was counted based on CD34 marked en- dothelial cells of micrevessels. Results:Survivin protein was undetectable in normal colorectal mucosa, while the ex- pression rates in adenoma and adenocarcinoma were 62.5% and 70.9% respectively. The expression of Survivin in adenoma and in adenocarcinoma was significantly higher than that in normal mucosa. MVD were 21.55 ± 10.61 in colorectal adenoma,28.89 ±10.47 in colorectal adenocarcinoma,8.23± 2.77 in normal mucosa. There were signifi- cant differences of CD34 expression in colorectal adenoma, colorectal ad , normal mueosa. The overex- pression of Survivin protein and CD34 was related to tumor depth and lymph node metastasis. The expression of Sur- vivin protein was correlated positively with MVD. Condusion:Survivin protein plays an important role in the onset and development of colorectal adenocarcinoma by inhibiting the apoptosis of cancer cells and inducing the proliferation of microvessels. Survivin protein and CD34 may be the parameters for prognosis in colorectal carcinoma.
